MESENCHYMAL STEM CELLS in 3D
- with and without RGD -
Human mesenchymal stem cells (hMSC) were encapsulated in degradable QGel™ matrix with (Figure 1) and without (Figure 2) integrin-binding RGD peptide. In
both matrix types, hMSCs viability was very high after 3 weeks culture (90-100%). Cell spreading was observed exclusively in the presence of the RGD peptide for cell adhesion.
MATERIALS
- Primary human mesenchymal stem cells
(hMSC, Passage 3)
- Approx. cell seeding density: 4 millions cells/mL gel
- QGel™ MT 3D Matrix: degr. with RGD (REF 1001)
- QGel™ MT 3D Matrix: degr. w/o RGD (REF 1004)
- Culture medium: DMEM + 10% FBS
METHODS
- Encapsulating cells & casting gel discs
- Cell viability assay

Figure 1: hMSCs are elongated and connected with neighboring cells when cultured for 3 weeks in degradable QGel™ MT 3D Matrix with RGD (REF 1001). Confocal image of hMSCs stained with Calcein AM for live cells (green) and ethidium homodimer for dead cell nuclei (red).

Figure 2: hMSCs remain round shaped when encapsulated for 3 weeks in degradable QGel™ MT 3D Matrix without RGD (REF 1004). Confocal image of hMSCs stained with Calcein AM for live cells (green) and ethidium homodimer for dead cell nuclei (red).
